研究概况 Summary of Sun Lab:

 

近5年发表SCI 论文55篇,其中第一/通讯作者SCI 论文35篇;第一/通讯作者影响因子累计超过100,单篇最高影响因子9.23,论文大部分发表在学科领域前20%的杂志。研究成果多次在Cell Cycle、Mol Hum Reprod等获得编辑和同行专家特邀评论、选为封面文章,被多本英文著作如Oogenesis收录并介绍;并在国际会议如ARBS上获奖及获邀WCRB等多个国际大会特邀报告。2011年国家优秀自费留学生。

研究方向 Research Interests:

  

   1. 配子生物学 Gamete biology

    本课题组的研究重点在卵母细胞成熟过程中的纺锤体组装,染色体分离和不均等分裂的机制。初级卵母细胞生发泡破裂(GVBD)后,染色质在卵母细胞中央凝集成染色体,微管在附近组装。微管组成纺锤体(Spindle), 染色体在卵母细胞中央排列。在微丝的牵引下,纺锤体和染色体移动到卵母细胞的皮层下,并由微管牵引染色体分离,通过不均等分裂的方式形成一个大的卵子和一个小的极体。纺锤体组装对于细胞周期的进行是必须的,染色体分离保证了遗传物质的稳定和准确性,不均等分裂的方式为受精和早期胚胎发育保留母系物质。因此,探清其调控机制对于了解生殖细胞的发育是极其重要的。课题组先前的工作证明WAVE2/JMY-Arp2/3-actin信号通路调节了卵母细胞的不均等分裂,这一通路已被收录入Oogenesis等英文著作中;近期的工作证明小GTP酶效应因子ROCK也参与了这一过程,并且其功能在小鼠和猪等哺乳动物中是保守的。

 

   2. 生殖毒理学 Reproductive Toxicology

   环境中存在的各种毒素及疾病能够显著影响哺乳动物的生殖能力。近年来国内食品安全问题频发,而女性流产现象日益突出,生殖相关问题日益增多;在家畜养殖中饲料毒素则造成家畜繁殖力下降、流产死胎等现象,极大影响了家畜生产的经济效益。对于人类及家畜出现的各种繁殖问题,卵子质量下降是重要原因之一。研究环境毒素或疾病对于卵子质量的影响,有利于为防治这一问题提供理论基础。近期实验室采用体内和体外模型,利用猪和小鼠两种哺乳动物,研究结果证明摄食含有霉菌毒素饲料的小鼠卵子质量下降,发育能力、亚细胞结构分布和定位均发生异常,同时DNA甲基化及组蛋白甲基化水平波动、氧化应激发生,可能是造成卵子质量下降的原因之一。
